Nope - all it needs is a list of blocks on the boot device, where to
load them, and where to start execution in the loaded code. The boot
loader contained in those block knows what kind of filesystem is present
on the boot media, and will locate, load, and execute a secondary boot
loader. The secondary boot loader can then locate, load, and execute the
specified kernel.
